Academic preconditions:
BM111 Eukaryot Molecular Biology (BM111) must be passed or followed simultaneously.
Course introductionThe objectives of the course are:
1) to give a theoretical introduction to the molecular processes leading to differentiation of eukaryotic cells (differentiation of adipocytes in vitro is used as a model system);
2) to give a practical introduction to several of the most commonly used techniques in molecular biological analyses of gene expression in higher eukaryotic cells. Furthermore, the course aims at giving the participants insight into how bachlor/ITEK/"speciale"-related research projects are planned and conducted.
The course is therefore a combination of lectures, presentations of relevant papers and intensive laboratory work where the students are actively involved in the planning of experiments.
Expected learning outcomeSubject overviewTheoretical introduction to adipocyte differentiation with special emphasis on transcriptional regulation.
Analysis of gene expression during in vitro differentiation of 3T3-L1 cells to adipocytes. This includes investigations of the expression of important transcription factors and adipocyte-specific genes.

Purification of RNA.
Northern blotting and hybridization.
Isolation of DNA fragments.
Multiplex RT+PCR
RNase protection.
Western blotting.
Electrophoretic mobility shift assay.

Expected working hours
The teaching method is based on three phase model.
Laboratory course (90 hours) and lectures and presentation of papers (30 hours). Intensive course, where long days (10-12 hours) can be expected some of the days.
