Course introduction
The purpose of the course is to give the student a introduction in basic protein chemical techniques which are used for charaterization of the primary structure of proteins and their identification.

Expected learning outcome
Statement of Aims:

• Describe the primary structure of a protein and explain the principles in the most importan methods for primary structure analysis
• Explain the principles in separation of polypeptides by HPLC
• Describe the principles in gel electrophoresis
• Explain the principles in amino acid analysis, Edman degradation and mass spectrometry of polypeptides, be able to analyze the related data and integrate these to solve the primary structure of a protein
• Explain the principle of a MALDI mass spectrometer
• Explain the principles in mass spectrometric protein identification and be able to analyze the related data.
• Explain the principles in mass spectrometric sequencing of peptides and analyze ms/ms data.
• To evaluate the techniques used in the course for analysis of protein structure.
• To use the computer programs available for the course  



Subject overview
The following topics will be treated during the course:
1. Primary structure of proteins
2. Chemical characteristics of amino acids
3. The principles of amino acid analysis and its use for
 a) Protein / peptide characterization
 b) Quantitation
4. Protein sequencing in the form of:
 a) Edman degradation
 b) Mass spectrometric sequencing
5. SDS gel electrophoresis of proteins
6. Liquid chromatography
 a) Reverses phase for peptide separation
 b) Ion exchange
 c) Solid phase extraction
7. Basic mass spectrometry (MALDI)
 a) Intact mass analysis
 b) Peptide mass fingerprinting
 c) MS/MS for identification
8. Bioinformatical protein analysis

Prerequisites for participating in the exam
None

Assessment and marking:
Project assignment. Pass/fail, internal examiner. Course report to be handed in three weeks after end of course.

If failed, the report can be resubmitted after two weeks
