Prerequisites:
None

Academic preconditions:
Bachelors degree in either Biochemistry and molecular biology or Physics and Chemistry. Due to instument availability a maximum of 12 participants can enroll in the course. For BMB students it is advised to have BMB509 passed

Course introduction
In the last years, a large number of state of the art experimental microscopy techniques have been developed. The main objective of this course is to provide the theoretical basis of these techniques together with some “hands on” laboratory practice.

Expected learning outcome
At the end of the course the student is expected to :

  • Evaluate scientific results in the original research literature
  • Present scientific results in an oral seminar given in English
  • Discuss strengths and limitations of modern techniques in microscopy
  • Identify the “method of choice” for analysis of a given biological sample
  • Perform image analysis using advanced microscopy instrumentation
Subject overview
Basic theory (including quantitaive image analysis) for the following techniques will be provided: 1) Laser Scanning Confocal Fluorescence microscopy 2) Spinning Disk Confocal Fluorescence microscopy 3) Total internal reflection Fluorescence (TIRF) microscopy  4) Multiphoton Excitation Fluorescence microscopy 5) Second Harmonic Generation microscopy 6) Fluorescence Lifetime imaging and FRET 7) Super resolution techniques (STED microscopy) 8) Fluorescence Correlation spectroscopy related techniques (FCS, RICS) 9) single Particle tracking (SPT)

Prerequisites for participating in the exam
None

Assessment and marking:
Oral exam, Danish 7 mark scale, internal examiner
Lab.work, pass/fail,internal evaluation by teacher
Remaks regarding evaluation:
(a) For the lab exercise, the students will be separated in 4 groups of 3 persons and work in teams in four different exercises (1 per group). Each group need to report and discuss their results to the rest of the class in a seminar. This activity will be evaluated, pass/fail grading by the teacher (1 ECTS);
(b) Oral examination. Grades by the 7 mark scale, internal examinator (4 ECTS).
